Comportements cellulaires et régulation génétique au cours des réactions d'immunité innée chez la moule Mytilus galloprovincialis ArchiMer
Li, Hui.
The mytiliculture is one of the most important aquaculture in the world, however, little is known on the mussel immune system, which plays a critical role in case of infectious diseases. Like all the other invertebrates, our model, the Mediterranean mussel Mytilus galloprovincialis, possesses only an innate immune system considered to be primitive and non-specific. In the present study, our results showed that the mussel immune system (i) is capable of responding differently to a variety of stimulations, suggesting a specific recognition process; (ii) reacts differently according to the geographic origin of the animals, showing an adaptation to their own environment; (iii) is influenced by the season, which explains the seasonal mortalities; (iv) presents...

Effects of dissolved mercury on embryogenesis, survival and growth of Mytilus galloprovincialis mussel larvae ArchiMer
Beiras, R; His, Edouard.
The effects of Hg concentrations ranging from 0 (control) to 1024 mu g l(-1) upon embryogenesis, survival and growth of Mytilus galloprovincialis mussel larvae were investigated. Embryogenesis was abnormal in 50% of the individuals at 10 mu g l(-1). The 48 h LC(50) for D-shaped, early umbonate, late umbonate and eyed larvae were 51, 164, 322 and 383 pg l(-1) respectively. The LC(50) was an allometric function of ash-free dry weight with exponent b = 0.60. Larval growth was significantly reduced after 6 d exposure to 4 mu g Hg l(-1) and after 10 d exposure to 2 mu g l(-1). No significant differences in lethal or sublethal sensitivity to Hg were found between M. galloprovincialis and previously studied Crassostrea gigas embryos and larvae. The implications...

Etude d'une prolifération hémocytaire chez la moule Mytilus edulis ArchiMer
Noel, Danièle.
Hemic proliferative disease in the bay mussel, Mytilus edulis, has been studied following mortalities in Washington State (USA). The disease has been reproduced in the labortory. The establishment of this experimental model led to study tumoral characteristics of the atypic hemocytes : ultrastrure, in vitro cultivation, transplantation. The infectious etiology of this tumor has been verified. It has been hypothesized to result from a viral infection since retrovirus-like particules have been observed following induction by bromodeoxyuridine. Specific monoclonal antibodies against atypic hemocytes have been obtained and characcterized

Fitness landscapes support the dominance theory of post-zygotic isolation in the mussels Mytilus edulis and M. galloprovincialis ArchiMer
Bierne, Nicolas; Bonhomme, Francois; Boudry, Pierre; Szulkin, M; David, Pierre.
We studied the genetic basis of post-zygotic isolation in the marine mussels Mytilus edulis and Mytilus galloprovincialis. Evidence was obtained for a high number of recessive Dobzhansky-Muller substitutions in the genome of these two mussel taxa. We analysed the segregation of unlinked diagnostic markers in the progeny of two backcrosses and an F-2 cross, 36 h and 200 days after fertilization. Directional selection favouring M. galloprovincialis genotypes was observed in both kinds of cross. In the F-2, epistatic interactions between each pair of chromosome fragments mapped by the markers were identified in addition. Our results imply that homozygous-homozygous interactions are required for breakdown of coadaptation, in accordance with the dominance...

Influence of temperature, salinity and E. coli tissue content on immune gene expression in mussel: Results from a 2005-2008 survey ArchiMer
Li, Hui; Toubiana, Mylene; Monfort, Patrick; Roch, Philippe.
Several bivalves, including mussels, suffered from mortalities particularly in summer. To look for the possible effect of environmental parameters on immune capacities, Mytilus galloprovincialis were collected monthly from August 2005 to July 2008 from the Palavas Laguna, French Mediterranean coast. Q-PCR was used to quantify the expression of three antimicrobial peptide genes (defensin, mytilin B and myticin B), in addition to lysozyme and HSP70. House keeping gene was 28S rRNA. Defensin, myticin B and lysozyme appeared more expressed in spring-summer than in winter. In contrast, HSP70 expression was higher in winter. Statistical studies using principal component analysis (PCA) and multiple regression models revealed positive influence of temperature on...

Karyotypic diversification in Mytilus mussels (Bivalvia: Mytilidae) inferred from chromosomal mapping of rRNA and histone gene clusters ArchiMer
Perez Garcia, Maria Concepcion; Moran, Paloma; Pasantes, Juan J..
Background: Mussels of the genus Mytilus present morphologically similar karyotypes that are presumably conserved. The absence of chromosome painting probes in bivalves makes difficult verifying this hypothesis. In this context, we comparatively mapped ribosomal RNA and histone gene families on the chromosomes of Mytilus edulis, M. galloprovincialis, M. trossulus and M. californianus by fluorescent in situ hybridization (FISH). Results: Major rRNA, core and linker histone gene clusters mapped to different chromosome pairs in the four taxa. In contrast, minor rRNA gene clusters showed a different behavior. In all Mytilus two of the 5S rDNA clusters mapped to the same chromosome pair and one of them showed overlapping signals with those corresponding to one...

Lysozyme gene expression and hemocyte behaviour in the Mediterranean mussel, Mytilus galloprovincialis, after injection of various bacteria or temperature stresses ArchiMer
Li, H; Parisi, M; Toubiana, Mylene; Cammarata, M; Roch, Philippe.
The aim of the present study was to evaluate the expression of the Mytilus galloprovincialis lysozyme gene in different in vivo stress situations, including injection of bacteria Vibrio splendidus LGP32, Vibrio anguillarum or Micrococcus lysodeikticus, as well as heat shock at 30 C and cold stress at 5 C. Injection of V. splendidus LGP32 resulted in: (i) a general down-regulation of lysozyme gene expression, as quantified by Q-PCR; (ii) reduction in the number of circulating hemocytes; (iii) decrease in the percentage of circulating hemocytes expressing lysozyme mRNA which was now restricted to only small cells, as observed by ISH; and (iv) accumulation of hemocytes expressing lysozyme in the muscle sinus where injection took place. Injection of V....

On the identity of broad-shelled mussels (Mollusca, Bivalvia, Mytilus) from the Dutch delta region Naturalis
Groenenberg, D.S.J.; Wesselingh, F.P.; Rajagopal, S.; Jansen, J.M.; Bos, M.M.; Velde, G. van der; Gittenberger, E.; Hoeksema, B.W.; Raad, H.; Hummel, H..
Late Quaternary (Eemian) deposits of the Netherlands contain shells that resemble those of living Mytilus galloprovincialis. Similar broad-shelled mytilids also occur in estuaries of the southwestern Netherlands together with slender individuals typical of M. edulis. We sampled living mussels along a depth gradient in the Oosterschelde to a) investigate whether a relation exists between shell shape and depth, b) test if the broadshelled specimens might represent M. galloprovincialis (or a hybrid with M. edulis) and c) assess by inference if the Quaternary specimens might be attributed to M. galloprovincialis as well. In order to do so, we compared genetic (length polymorphism of Me 15/16, COIII sequences and AFLPs) and shellmorphological characteristics...
